"""Second cancers are a leading cause of morbidity and mortality among cancer survivors. Childhood cancer survivors are particularly vulnerable to the induction of second cancers associated with radiotherapy and cytotoxic chemotherapies used in first primary cancer treatment. Although ionizing radiation and alkylating agent-based chemotherapy are known carcinogens, the biologic mechanisms underlying the carcinogenic processes are unknown. Recent advances in technologies for characterizing the mutational spectrum in archival tumor tissues coincide with the maturation of several long-term follow-up studies of cancer survivors with detailed data on treatment exposures, providing a unique opportunity to discover the biologic basis of treatment-induced second cancers.
